14 3 3 raf. Phosphorylation events play a crucial role in raf activation. We propose that dimeric 14 3 3 is needed both to maintain raf in an inactive state in the absence of gtp bound ras and to. In addition the antibody reacts with a synthetic phosphopeptide corresponding to the 14 3 3 binding site on raf at s621 but not with the unphosphorylated peptide or with a phosphopeptide from a non 14 3 3 binding region of raf figure 1e compare lane 2 with lanes 1 and 3. More than 200 signaling proteins have been reported as 14 3 3 ligands.



Inhibitors are effective against the mutant b raf but paradoxically they activate wild type b raf limiting their therapeutic potential. Many human melanomas contain an overactive form of raf kinase b raf. In general it seems that interaction of 14 3 3 with ifs interferes with other effector pathways.



We propose that dimeric 14 3 3 is needed both to maintain raf in an inactive state in the absence of gtp bound ras and to stabilize an active conformation of raf produced during activation in vivo. Also 14 3 3 interacts with phosphovimentin displacing the 14 3 3 partner raf which can no longer be activated by egf tzivion et al 2000. This gene product belongs to the 14 3 3 family of proteins that mediate signal transduction by binding to phosphoserine containing proteinsthis highly conserved protein family is found in both plants and mammals and this protein is 99 identical to the mouse and rat orthologs.



The 14 3 3 protein family has received considerable attention recently in the literature because of the finding that β and isoforms interact with and activate raf. Ras gtp displaces 14 3 3 from raf 1 but s259 is still phosphorylated. 14 3 3 proteins are a family of conserved regulatory molecules that are expressed in all eukaryotic cells.



Phosphorylation of serines 259 and 621 in c raf and serines 364 and 728 in b raf has been suggested to be critical for association with 14 3 3 proteins. Determined the structure of a phosphorylated b raf dimer in complex with the scaffold protein 14 3 3 by cryoelectron microscopy.
